I included the feature as part of Fluffy Manager (mod manager and trainer for various RE games). How to enable coop: change current game to Revelations 2 in the menu, launch Revelations 2, and then turn on local coop (have to activate trainer first), and then choose a chapter in the game or restart checkpoint, and that should do the trick.
There's a couple of known issues:
- Keyboard controls affect both players (I doubt I can fix this).
- Raid mode local coop doesn't work (hopefully I can get that working).
- If you turn coop on/off while you're switching between Claire/Moira in singleplayer mode, the game might get really confused. Toggling coop on and off works best while the game is paused.

That sounds like they'll consider adding local coop in a patch. I'd almost expect them to since it would be really easy for them to do so.
