I think its ~16GB sized data.

I need the USB stick, and I'd like to back up the data on my computer.

I've tried the Disk Manager in Windows, and MiniTool Partition Wizard, but both of them are reading the USB as 'unallocated' and/or 'empty' and won't let me copy the data.

MiniTool won't let me clone the USB either, as it says the USB is empty.

Is there any way to force a copy or disk cloning function?

Maybe try creating an image with Clonezilla?

The drive apparently has DRM to it so it's a bit weird.

A quick google search took me to a OperationRainfall page to get around the DRM but i'm not sure if it'll work for what you're doing.

Just plug another storage medium into a USB slot and make a copy of it, I'm afraid.
