Hmm lets see. First thing I'd do is ditch the Pokemon. They feel horribly out of place and Pokken exists now. Then after that.

I don't know, I think there are a lot of characters Nintendo can add. It's just that aside from the Inklings there aren't going to be as many A-listers left.
I tried to compile a list of the people NeoGAF voted for back when the Smash 4 ballot started. Didn't keep track of numbers though, but I think the most popular ones were pretty obvious: Toad and Inkling and the like.

What we know so far:
Let me emphasize that what we already know is basically rumors from reliable source(s). We don't have any idea on what to expect from Sakurai, Nintendo and Smash NX, other than the game is planned to be one of launch titles for upcoming console, NX.
The initial tweets regarding Smash NX came from one of the most reliable sources, known as Dr. Serkan Toto. He has been correct regarding numerous Nintendo rumors as far as I know. He broke the initial rumor on January 28, 2016.
Another source is Emily Rogers. I'm not sure if she heard the rumor from her own source, or just repeated what she heard from Dr. Toto. Hopefully we will get some kind of clarification on that.

To answer your question about characters. There are still characters that people wanted to see in Smash Bros:
Inkling
Rhythm Heaven
Wolf
Ice Climbers
Snake (low chance)
7th Gen Pokemon (like Robin Hoot, Robin Root, or final form of Grass Owl)
King Rool
Dixie Kong
and many more.
He could've put something like "Game" or "What if". The title makes it initially seem like an official announcement of some sort has been made.

2. Luigi - Kind of a "waste of space" but he's basically mandatory
3, Yoshi - Maybe not 100% needed but I don't think it'd be wise to drop him
4. Peach - Not in the original but she's impossible to drop now
5. Bowser - Both a heavyweight and villain
6. DK - Basically mandatory
7. Link - No shit
8. Zelda - Maybe make her down B turn her into Sheik again?
9. Samus - Obvious
10. Fox - There would probably be riots if he weren't in
11. Captain Falcon - Can't let him go~
12. Pikachu - Basically mandatory
13. Mewtwo/Lucario/some other fan favorite - Don't know enough about modern Pokemon to know who is most worthy
14. Kirby - It's yo boi
15. King Dedede - Another heavyweight and villain slot

The only charcter that looks out of place in Smash is Ganondorf. And that's because of the Art style. He looks straight up goofy.
The art style in Melee and Brawl suited him more.

Starting from zero & rebuilding the gameplay from the ground-floor in a way that can better satisfy Melee fans as well as existing Smash 4 players (not to mention introducing new mechanics that can heavily change how Smash is played, like replacing Final Smashes with a dedicated Super Meter). It's the same reason why Capcom, Arc System Works, or SNK start from scratch for new numbered entries of their fighting games.

Honestly, would they really have to build a whole new engine for that?
The Project M team was able to successfully radically overhaul Brawl's engine into something that was basically identical to that of Melee's. I don't see how an official dev team with 100x the budget and development power couldn't do that, provided they even wanted to make a Melee styled game.
