Soul Reaver's intro. That intro blowed my mind away. Another aspect that blew me away from was plane shifting, I'm amazed that no other game tried such a thing. At least that I know of.

When Shadow of the Colossus started I immediately knew that it wouldn't be just another game.

And of course, when I heard "Weiss, you dumbass!" for the first time I knew I was in for a treat.

Fighting the Metal Gear basically immediately in Metal Gear Rising Revengeance made me know that it would be an incredible game.

Near the beginning of Dark Messiah when you wake up with an assassin in your bedroom. I kicked him down the stairs. Then he groaned in pain and started climbing up the stairs. So I kicked him down again.

That alone taught me everything I needed to know about the game.

The opening chopper scene in FarCry 3: Blood Dragon.

Just instantly made me think "this game doesn't mess about!"

Walking on the Presidium in Mass Effect 1 and learning about all these different species, cultures and civilizations. It felt like I was taking my first steps into a much larger world.

Dante swinging an enemy into the air and holding them there with his pistols in the first Devil May Cry.

It's still cool, but in 2001 there was nothing cooler.

In the first few seconds of Megaman X, as you appear, vehicles are moving past you, fleeing the enemy robots and trailing smoke. I always thought that was a great background touch that said 'the city is under attack' in a few seconds without any words.
 
Opening few minutes of Deadly Premonition. I didn't know a single thing about the game going in, and the strange style with which the first cutscene was directed, York's bizarre diatribe about Tom & Jerry, the mystery as to who the hell Zack was, and the immediate plunge into a supernatural otherworld had me completely enthralled from the instant I began.
 
Seeing the Hyrule field for the first time in Ocarina of Time.


Jumping into the devastated overworld in Terranigma.


Teleporting to 600AD in Chrono Trigger.

Kicking the can in Mega Man Legends.

Honourable mention to the area immediately after that, where you can jump on top of a truck, be driven around for a while, use the height of the truck to make the jump onto the roof of a nearby house, and then jump from roof to roof.
Actually, the first boss, too -- just how huge and intimidating he was. Really, the whole game is made of moments like this.

The moment you got Epona for keeps in Ocarina of Time. When you exit the ranch and ride around Hyrule field for the first time... magic.

Walking through a portal made from the portal gun in Portal. I was at a friends house and he showed me the game that's why it was not directly at the beginning.

Summoning and invasions in Demon's Souls. I was scrapped out the gate in 2-1.

Being able to play with gravity on individual planetoids in Mario Galaxy renewed my love for gaming.

Learning that you can open the cinema door in the first level of Duke Nukem 3D. I hadn't seen anything like that before, and it totally blew my mind.

Another thing was flying for the first time in Child of Light. I was loving the setting, art style, combat system and writing so far, but after acquiring this ability I realized this game was a masterpiece.

Oh, and let's not forget about Ace Attorney 1. I was skeptic about it at first, but decided to try it anyway. Can't remember the exact point it happened, but at the end of the first case one thing was clear: this was awesome.
 
Akira kicking ass in Virtua Fighter, low res polygons and all.

Wavy time travel transition in Chrono Trigger.

Leaving the shelter and seeing the sun for the first time in Fallout 3.

Bayonetta slowmotion dodge and all the graphical effects surrounding it.

The start of MGSV. You go through the tiring, exciting, long opening sequence then Ocelot starts explaining the situation to you. Then this song starts playing and tells you "yep, there's fun times ahead!"

Too bad the game as a whole didn't meet such lofty expectations.
