In Mario Kart because the Kart is part of the player, the mushroom is fed to the Kart which means that the mushroom reacts along with the petrol as part of the combustion thus creating a small explosion which propels the character along at a slightly faster speed for a short period of time.

It's likely reasonable to assume that Super 'Shrooms, on a basic level, are a potent source of energy. In a living organism, the user experiences a rush that can do anything from restoring health (especially in the RPGs) and/or increasing power and/or mass.

Meanwhile, in a machine such as a Kart or Bike, the energy mixes with the fuel in the machine in order to make for a reaction not unlike a nitrous oxide boost.
