It was a quintessential RPG starter village, as if all the starter village tropes of the 8 and 16 bit era were distilled into a single map.
 
I agree with almost everything mentioned here, so I'll add some that haven't been touched on yet:

Final Level. The music. Everything.

Also, this game had two intro stages, technically, and Alucard's was just as cool, if not better. Richter's set the tone for the rest of the game perfectly, though.

Wow, gamers indeed.

I gave a presentation about a game idea once. I said something along the lines of the battle system resembles Chrono Trigger and some other game mixed together. Actual developers from game companies asked me what Chrono Trigger was...... I was stunned.
 
Rez -- Area 1 just sets the tempo for the greatest hour of your life. It's still possibly my favorite of the 5 levels. I had bought the game on a lark on PS2, and to this day it's one of my top 3 games of all time, if not número one.

Also, I don't know if it really counts or not, but Pac-Man CE...? All I know is that I downloaded the demo just to check it out, and when it faded out after 3 minutes to the unlock screen I hammered the "BUY" button as fast as I could to unlock the full version.

The opening of Ar Tonelico 1 only takes place in a small portion of the tower, but the tower as a whole is quite probably the most memorable piece of gaming architecture I've come across.

Other notables include Above the World from Takkoman (dat music) and the Hydra battle from WO3 (dat hopelessness).

Azuremyst Isle from WoW. First character I ever made was a Draenei and it's still the only place in the game that gives me the nostalgia chills. Everywhere else has been changed pretty heavily.

Elwynn Forest should be memorable to anyone who's played WoW. For some reason it's always really populated compared to the Horde areas, they don't really have a similar place.

It's the closest thing to the old Star Wars Galaxies cantinas in WoW.

Draenei a first is a rarity, and yeah, the BC areas must feel like islands of greenery in a sea of urbanization by now.

Mine was Tirisfal Glades. Felt like wrong shit went down everywhere you looked. Fit the Forsake perfectly. Do not go past the Bulwarks.

Thinking back, it reminds me just how on point Blizzard used to be about guiding with visual cues and drawing attention, and teaching via the player doing. They've degenerated badly in these reguards...
 
Frigate Orpheus (Metroid Prime) and the descent into Rapture (Bioshock) are what immediately came to mind for me.

Loved the lighthouse in Chaos Theory. Dark, bleak, wet, creepy.

(and Sam's face was nightmare fuel in this game, making it even creepier)

Has to be God of War 3 for me. Staggering introduction to a game. I hadn't played any of the prior GoW games either so I was unaware largely of how epic and expansive Santa Monica made the franchise.
 
While we're talking WoW starting zones, i like Gilneas. Too bad it's as dead as a zone can be once you're done with it. It's even more dead than Silithus. Such wasted potential.

Was playing this when we got a NES after playing Atari and Commodore64 games for years - Completely redefined what 2d platforming was all about. Also a perfect example of how to introduce a player to gameplay without blabbering too much, the player just finds out how to interact with blocks and coinBlocks as well as the goombas by himself.

Shinjuku Hospital in SMT3. Extremely atmospheric and creepy level set the tone for the rest of the game and I knew right off the bat the game was going to be something special.

Space Frigate Orpheon in Metroid Prime 1, quite simply the best tutorial level ever.

(more like, first hub area if you roll a Hobbit...but still)

And The Shire is my favorite zone of any MMO. It was so charming and really felt how I've always imagined the Shire would. The quests were great, too, fitting in with the theme of the zone perfectly.
 
Mentioned already, but another shout out to Metroid Prime's Frigate Orpheon. To this day it remains the best opening stage/area that I've experienced. Wandering through a deserted derelict ship, piecing together what happened from the visual cues and the scanning logs, then an escape sequence. So good.

A close runner up is Black Mesa in the original Half Life. Starting off going through the facility while it is still normally functioning and staffed, only to watch it go straight to hell.

God of War-The Thread.

All 3 console games had mindblowing and riveting beginning levels for their respective time. 3 is still amazing since we're still in this generation.

A lot of talk about the WoW zones. Yes Elwynn forest left a mark on me in 2005 when it was released in europe and playing as a human warlock. And off course going through the dark portal for the first time in TBC and seeing a alien world. Good memories of that one.

And also Unreal when you go out of the ship the first time with 3dfx very mind blowing in the 90's.

GOW 3 - There is nothing that I have played in all my years of gaming that comes remotely close to the epicness that is the first 30 minutes of GOW3.

Clock Town is one of the best levels with so much to do. The music is great, the side quests flesh out the people in town and the knowledge of only having 3 days left to stop the moon was amazing.
