Also the main composer from the Japanese branch of Castlevania is in Smash 4, it should be a nice opportunity for giving Yamane one last change to work with the series that made her famous.

Actually, it's fairly likely that Yamane will continue to contribute music to future Castlevania entries regardless - she's worked on the soundtrack for the last 3 Castlevania pachinko machines, and is probably attached to the current one as well.

I think Smash should keep Nintendo as its focus, and I find people's obsession with third-party characters a little strange. I fully welcome the characters they have chosen so far because they're all iconic and important to the entire industry. That's why I'd love to see Snake back too. He's not just a random third-party character like, say, Shovel Knight. I love Shovel Knight, but including him would cheapen the roster to me. You don't just get into Smash. Especially not if you've only been in one, multiplatform game.
That said, if they were to add more non-Nintendo characters, who should they go with? Other than Snake, they have all the biggest icons covered already. Look up any "most iconic game characters of all time" list. Sonic, check. Pac-Man, check. Is it time for us to lower the bar just a little bit and allow smaller characters to get in? Personally, I think we have enough third-party characters for now, but for the sake of this argument, let's just say yes. I like the idea of having the biggest Japanese companies represented in Smash. If they were to go for that, Level-5 definitely deserves representation, and Professor Layton would be my personal choice. It's a shame Square Enix don't really have an iconic and recurring mascot that would be fun to play as, because they totally deserve a character. If I could have it my way I would bring in a Konami character, because that's a huge void they haven't filled yet. Bringing back Snake or introducing Simon Belmont - both sound like great ideas to me.
I recently played the Layton series and I absolutely love it. It's a great world with some great characters. If they were to add him to the game, I'd most likely enjoy his inclusion and add a bunch of new songs to my favorites in Sound Test mode. However, I do think it would be weird seeing him next to Sonic, Mega Man, Ryu and Pac-Man. I'll take him any day over Rayman and Shovel Knight though.
Rocket Slime was on DS, you're thinking of it's Japan only prequel on GBA. I'd also say Rocket Slime is pretty easily the best Zelda game on DS.I played it when the first came out for the GBA. An obscure game about slimes piling up and charging forward. I got bored fast.
As far as cute RPG blobs go, it does nothing special apart from combining into a King Slime. Dragon Quest fans want to play with the main characters, not with the monsters.

Simon Belmont really isn't that iconic. The only truly iconic Castlevania protagonist IMO is Alucard (though Shanoa is my favorite).

Square has Chocobo, Moogle, maybe Cactuar, Tonberry and..... Well, Cloud, but that's never been a part of Nintendo's history. Even less than Snake.The first 4 could have interesting movesets, though.

I wouldn't say a Rhythm Heaven rep is as guaranteed as the Inklings. But seeing as we almost got one for Smash 4 (& how big RH is overall), I wouldn't count a playable Rhythm Heaven rep out period. For all we know, Sakurai could be finishing up said Rhythm Heaven rep to be Smash 4 DLC. Simon is the first and most recurring Castlevania protagonist. He was in Captain N, DreamMix TV World Fighters, and Konami Wai Wai World, and he's the top billed character in Castlevania Judgment (over Alucard). Simon is far and away the most iconic Castlevania character.
