Cave Johnson is the funniest by a country mile.

Yep pretty much my answer.

One of, if not THE funniest character in a game that I've played. Everything he said was funny. I mean Glados was funny too and so was Wheatley but none of them made me laugh as much as Cave Johnson. Having J.K Simmons voice him too was just perfect

Gotta agree with OP on HK-47 being up there. That being said, Jolee Bindo from the same game gives him a run for his money. Also, see Black Whirldwind from Jade Empire and quite a few Borderlands Characters.

Oh yeah, CL4P-TR4P went from insufferable and Jar-Jar level of annoying in Borderlands, to genuinely amusing (for the most part) in Borderlands 2. I have no idea how it happened, but it did. o_O

Personally dislike Jak series, its not the type of platformer i enjoy. But Daxter was the one reason to play that game, he was funny and Naughty Dog really made a funny character that never overstayed his welcome.

Every single line from Handsome Jack is pure gold. Borderlands 2 in general has some of the funniest dialogue in all of gaming.

Really like Daxter and Cpt Qwark, too.

Sam and Max in Sam & Max Hit The Road. Timing could be a bit off though, since sound mixing wasn't up to par yet back in those days.

Other than that, Portal 2 seems to be the only game I've played written by actual comedy writers. In games like Borderlands and Blood Dragon they are trying too hard most of the time.

Oh, Mr. Torgue is hilarious too, in both the Torgue and Dragon Keep DLCs. He never fails to crack me up, and the Fake Geek Guy gag in Tiny Tina's DLc was brilliant.
